If it boots on a UEFI PC then it could look in either of the boot or the EFI directories and end up being told to go to the same grub.cfg file for the detail boot instructions. If it boots on a MBR PC then it will look at the menu.1st file for further instructions and there told to look into that grub.cfg file which contains the detail boot instructions. In the partition you will see many files and folders. Setting both those flags means that on whatever computer it boots on with whatever it is set to boot with in the shape of UEFI or MBR the boot process end up looking in that partition for further instructions. You will also notice that there is both the boot flag and the esp flag set. Thus for this Linux on this USB only need one FAT32 partition was needed. Recent Windows wim file size is bigger than 4 GB and that needs a NTFS partition as well as the FAT 32 one with a reference to it during the boot process. With the GParted you will see that for this Linux Distro (Puppy) I did not need to bother with a partition and a file system that copes with files larger than 4 GB because there isn't any.
